Spanning four decades, Castelli Graphics contributed significantly to an increase in popularity, technique and appreciation of fine art printmaking in the United States. Maintaining a regular exhibition schedule, the gallery also published or co-published many editions of prints by significant artists of the Twentieth Century. Artists whose work was exhibited at Castelli Graphics included Richard Artschwager, Joseph Beuys, James Brown, Dan Flavin, Jasper Johns, Donald Judd, Ellsworth Kelly, Joseph Kosuth, Roy Lichtenstein, Bruce Nauman, Claes Oldenburg, Robert Rauschenberg, James Rosenquist, Ed Ruscha, David Salle, Richard Serra, Frank Stella, Bernar Venet, Andy Warhol.

In addition, the gallery exhibited and promoted the careers of many fine art photographers of the 1960's, 70's and 80's, including Robert Adams, Eve Arnold, Lewis Baltz, Ralph Gibson, Diane Keaton, Mary Ellen Mark, Peter Mauss, Sandy Skoglund, Eve Sonneman, Judy Tomkins.

Antoinette (Toiny) Castelli opened Castelli Graphics at 4 East 77 Street, New York City, in 1969. The gallery remained there until 1988, the year after her death, when it moved downtown to Soho. Castelli Graphics Soho, at 578 Broadway, opened in 1988 and closed its doors in 1997.
